A church dedicated to Saint Chiara is mentioned on a map of Ripatransone as early as 1598, and it was rebuilt between 1750 and 1754. It was consecrated immediately following this reconstruction by Bishop Niccolò Recco.
During the period of Napoleonic rule, the church and convent, like many other places of worship in Italy, were suppressed, and the Poor Clares were forced to relocate. A few years later, the buildings were purchased by Bishop Ludovico Luigi Ugolini, who transferred the parish church of San Benigno there. Subsequently, the church became part of the episcopal seminary.
From the second half of the 20th century onwards, the entire building underwent major restoration work; the decorations were restored, and the high altarpiece, a work by Michelangelo Bedini, who also painted other frescoes in the church, was installed.
Later, without being deconsecrated, the building became a venue for cultural activities and even served briefly as a diocesan museum.
